In vitro regeneration using meristem explants to induce shoots and roots in Mucuna pruriensrngenotypes were investigated. Different treatment combinations of culture media containingrngrowth hormones, nutrients and vitamins were evaluated to promote shoots and roots directlyrnfrom the meristem explants and indirectly via callus phase. A total of 13 experiments werernconducted. Among the various cytokinins, BAP, Kn, 2iP, GA3 and TDZ tested, the survivalrnof tissue (shoots and callus) was found to be effective in BAP combined with an auxin NAA.rnThe cytokinins, BAP and TDZ induced multiple shoots. An optimal level of 0.1mg/l BAPrnsupplemented with liquid media containing ¼ G-B5 + 7.5 g/l sucrose supplemented withrn5mg/l IBA was effective in root induction. BAP along with an auxin NAA was effective forrnsurvival of tissues (shoots as well as callus). Rooting was induced at the base of shootsrnproliferated on media containing lower concentrations of BAP. However, shoots proliferatedrnin TDZ containing media were difficult to root. No organogenesis was observed from callusrnregardless of the different media evaluated. Among the accessions, 10084, 14880 and 15169,rnaccession 14880 performed relatively better in tissue survival percentage as well as percentrnof shoot survival.
