Securidaca longipedunculata Fresen, locally called Etse-manahi, is semi-deciduous shrub or small tree that belonging to the family Polygalaceae and found in rainfall andrnequatorial humid areas of Africa. It is an indigenous medicinal plant in Africa and hasrnimportant place in the traditional medicine as well as modern medicine. This plantrnbecomes endangered because of high seed dormancy, low germination rate, overrnexploitation and long growth period. Therefore, micropropagation method is importantrnto address these problems. Thus, the objective of this study is to developrnmicropropagation protocol for S. longipedunculata from shoot tip explants. Seeds were de-coated and disinfected using 3%, 5%, 10% and 15% (v/v) Clorox and cultured on growth regulators-free solid MS medium. Shoot tips were initiated using variousrnconcentrations of 6-Benzyl Amino Purine (BAP) in combination with Thidiazuron. MSrnmedium containing different concentrations of BAP in combination with IBA; TDZ inrncombination with IBA or NAA were used for shoot multiplication. Shoots were culturedrnon full and half strength MS medium containing IAA, IBA or NAA for root induction. Thernhighest percentage of shoot initiation (87%) was obtained on MS medium containing 1.0rnmg/l BAP. Maximum number of shoots (8.5 Â± 0.69) was achieved on MS multiplicationrnmedium containing 1.5 mg/l BAP in combination with 0.1 mg/l IBA. Shoots gave highestrnnumber of roots (3.73 Â± 0.69) on full strength MS rooting medium containing 2.0 mg/lrnIAA.
