Background: Most malaria control strategies depend on safe and effective drugs, although, drugrnresistance is the most serious problem in achieving control of malaria. Artemether/Lumefantrinern(coartum®) has been used as a first-line treatment for uncomplicated Plasmodium falciparumrninfection since 2004 in Ethiopia. The spread of Plasmodium falciparum resistance to almost allrnavailable affordable mono-therapy, in many malaria endemic regions, is a serious impediment onrnmalaria control. As a result there is a need to a periodic evaluation of efficacy of antimalarialrndrugs.rnObjective: The aim of this study was to evaluate the efficacy and safety of Coartem® for therntreatment of uncomplicated Plasmodium falciparum infections in patients attending Gende WuharnHealth Center Gondar Zone, North-West Ethiopia.rnMethod: One arm prospective in vivo evaluation of the efficacy of artemether/lumefantrine (AL)rnwas conducted in patients with uncomplicated Plasmodium falciparum infection. Standard sixrndose regimen of AL was given twice daily for 3 days under supervision and clinical andrnparasitological responses during a 28 day follow-up period were assessed using a standardizedrndrug efficacy record form, on days 0, 1 ,2, 3, 7, 14, 21, 28 and adverse drug events as well asrnHgb changes were also investigated.rnResult: A total of 81 P. falciparom mono infected patients were included in the study, majorityrn82.4% were males. Overall, the 28-day clinical and parasitological cure rate was 98.8% (95%rnCI=93.3-100%). The mean temperature and parasite count at enrollment were 37.9°C andrn13,441.6rings/μL, respectively. Gametocytes were detected in 4.4% of the study participants onrnthe day of inclusion and the mean hemoglobin level was 13.7 g/dl. Arthemesnin basedrncombination therapy treatment rapidly cleared parasitaemia and fever by day three and completerngametocyte clearance was obtained on day 21. Insignificant hemoglobin recovery was observedrnamong patients with adequate clinical and parasitological response. There was no severe adversernside-effects were observed among these patients. Conclusion: Artemether Lumefantrine wasrnefficacious for the treatment of uncomplicated Plasmodium falciparum malaria in Gende Wuharntown.rnKeywords: P. falciparum, Coartem, ACT, Efficacy, Gende Wuha, Ethiopia
